Optimal pH & concentration of Histidinol Dehydrogenase on mutated C154A and H262N HisD plasmids.
Histidinol Dehydrogenase is an oxidoreductase that catalyzes the NAD-dependent oxidations of L-histidinol to L-histidine. The purpose of this experiment was to analyze Histidinol Dehydrogenase in vitro by assessing the optimal enzyme concentration and the optimal pH. This was achieved by performing a NADH assay to analyze the enzyme. The optimal enzyme concentration was found to be between 1-3μL which was the same range found in other literature results for HisD in P. pastoris. The optimal pH was found at a pH of 9.5 which again is around the same as other literature experimental results found for HisD in E.coli.
